摘要 基因-放疗是将放射敏感性调控序列与治疗基因和药物相耦联,在肿瘤放疗同时诱导目的基因和药物在肿瘤内高效表达,从而提高肿瘤放射敏感性,它是近年来肿瘤放疗新策略\[1\]。早期生长反应-1(early growth response-1,Egr-1)启动子在电离辐射诱导下能增强下游基因表达,并可提高放疗效果;乏氧反应元件(hypoxia response elements,HRE)在乏氧条件下可增强下游基因表达水平。笔者利用HRE和Egr-1构建乏氧放射双敏感嵌合性启动子HRE/Egr-1诱导寡核苷酸药物Dbait表达,并观察该重组质粒在乏氧条件下对人宫颈癌Hela细胞的乏氧放射增敏效应。
